We appreciate the comments of Drs Formiga, Ashton-Prolla, and Achatz in their letter addressing our article, “Earlier colorectal cancer screening may be necessary in patients with Li-Fraumeni syndrome.”1 The authors report that, in their cohort of TP53 carriers, colorectal cancer (CRC) rates were similar to those rates reported in our analysis, and they also notably observed a CRC diagnosed before the age of 25. However, the authors also importantly comment that CRC was rarely seen in patients with Li-Fraumeni syndrome (LFS) with the Brazilian TP53 founder mutation c.1010G>A (p.Arg337His, R337H).
